In the place of the immunenzymatic reaction, a colour spot emerges. It is assumed that one spot denotes one cell secreting protein of interest. In the case of rapidly dividing cells, or when the cells are cultured over a longer period of time, the term "spot forming unit - SFU" seems more appropriate. On the left-hand side, ELISpot results from a culture with majority of cells secreting protein of interest (in this case interferon-γ).
